Yes, in-place upgrades from 2012 R2 → 2016 → 2019 are supported. However, always backup first and test. Clean install recommended for Hyper-V hosts to avoid driver/configuration issues.
2016 introduced major changes: Shielded VMs, Storage Spaces Direct, Nano Server, Software-Defined Networking, Containers, and Windows Defender ATP. 2012 R2 is simpler and works for basic file/print/AD roles.
If you run more than 2 VMs per host, Datacenter is cheaper. For 5+ VMs, Datacenter pays for itself. For 1-2 VMs, Standard suffices. Consider your virtualization density carefully.
Unlimited Windows Server VMs on that host as long as you have hardware resources (CPU, RAM, disk). You can run dozens of VMs per physical server with Datacenter license.
